What is Lymphocyte Activated Cell
Therapy (LAK)?
Lymphocyte Activated Killer (LAK) Cell Therapy is a cellular immunotherapy that enhances the cytotoxic potential of the patient’s own immune cells. Lymphocytes, primarily T cells and natural killer (NK) precursors, naturally patrol the body to detect and eliminate abnormal or malignant cells. However, in cancer patients, tumor-induced immune suppression often weakens this defense. LAK therapy addresses this by isolating lymphocytes from the patient and activating them ex vivo with interleukin-2 (IL-2), transforming them into highly cytotoxic effector cells capable of overcoming tumor resistance mechanisms.
How the Therapy is Developed
The treatment process begins with leukapheresis, a technique that separates lymphocytes from the patient’s blood. These cells are transferred into our cGMP-compliant, ISO-certified immunotherapy laboratory, where they are cultured with high-dose IL-2. This cytokine stimulation induces proliferation and functional activation, converting quiescent lymphocytes into Lymphocyte Activated Killer cells. Activated LAK cells demonstrate increased cytotoxicity against a wide range of tumor cells, independent of antigen presentation, while expanding significantly in number to amplify their therapeutic impact upon reinfusion.
How it Works in the Body
After activation and expansion, LAK cells are reinfused intravenously into the patient. Once in circulation, they traffic to tumor sites and exert direct cytotoxic effects through perforin- and granzyme-mediated apoptosis of malignant cells. Unlike antigen-specific T cells, LAK cells can lyse tumor cells without requiring prior recognition of a defined tumor-associated antigen, giving them broad-spectrum anti-tumor activity. Additionally, their release of cytokines, including interferon-gamma (IFN-γ), modulates the tumor microenvironment, enhancing antigen presentation and stimulating other immune effector cells.
The Synergy of LAK and Dendritic Cell Therapy
When combined with Dendritic Cell (DC) Therapy, LAK therapy provides a complementary mechanism of action. DCs prime and expand tumor-specific cytotoxic T lymphocytes by presenting processed tumor antigens, creating targeted immune surveillance. LAK cells, in contrast, provide immediate, non-specific cytotoxic activity against tumor cells, including those that evade antigen recognition. Together, the two therapies generate both targeted specificity (via dendritic cell–induced T-cell activation) and broad killing capacity (via LAK cytotoxicity). This synergistic effect not only enhances initial tumor clearance but also promotes long-term immune memory and reduced relapse risk.
